Overview
Product & regulatory status
Investigational compound with FDA-identified safety concerns.
Endogenous hormone stimulation
The compound enhanced pulsatile GH secretion in the trial, rather than replacing GH directly. [1]
What the evidence shows
Older-adult study
A randomized trial in 65 healthy adults aged 60–81 found increased GH, IGF-1, and fat-free mass. The increase in fat-free mass did not produce measurable improvements in strength or function. [1]
More mass did not mean fat loss
The study did not show a significant advantage in visceral fat or total fat mass. Body weight increased, and fat-free mass includes water as well as tissue. [1]
What remains unknown
Laboratory hormone levels and lean-mass measurements cannot alone demonstrate improved independence, athletic performance, or longevity.
Safety
Metabolic tolerability
Fasting glucose increased and insulin sensitivity decreased. Increased appetite, leg swelling, and muscle pain were among reported adverse effects. [1]
Heart-failure signal
FDA highlights a hip-fracture trial stopped early because of a potential congestive-heart-failure signal. [2]
Oral research formulation
The study used controlled oral trial supply. It does not verify the contents of supplements or research products sold as MK-677. [1]
Understanding product quality
Compounded drugs are not FDA-approved finished products. Poor preparation can cause contamination or incorrect strength. A catalog entry or a research paper is not a quality assessment of your product. [3]
When to seek urgent help
Severe allergic symptoms such as trouble breathing, throat or tongue swelling, or fainting need immediate help. Call 911 or your local emergency number. This is general emergency information, not a complete list of risks for this compound. [4]
A starting point for discussion
This is not a complete list of adverse effects or interactions. Your health history, other medicines, and the exact formulation matter. Missing safety data is not evidence of safety.
Common questions
Does “nonpeptide” mean it is a dietary supplement?
No. Chemical size or structure does not establish supplement status, approval, or safety.
Questions for your clinician
Does the evidence match my condition and this exact product? What risks or interactions matter for me? How would we assess whether it is helping?
